The Class 721 diesel locomotive with electric power transmission, designed for shunting service and light line work, was manufactured between 1962 and 1972. The locomotive is derived from the ČME2 series, intended for export to the USSR, with both designs based on the T435.0 series (later redesignated as 720). The locomotives proved very successful, being quite popular and reliable in operation.

The power unit (type 6 S 310 DR) corresponds to the original T435.0 series, with the same power output. The locomotive has a longer frame, rocking bogies, and a larger fuel tank (4,000 l). The maximum speed increased to 80 km/h, and modifications to the TE 004 type traction motors resulted in an increase in continuous tractive effort to 12 tons.

After approximately 20 years of service, the first locomotives began to be withdrawn due to obsolescence and inefficiency – one of the successors was, for example, the Class 740.

A total of 290 units of Class 721 were manufactured.
